Excessive bubbles in SDS-PAGE?
There's an incident which I used a purchased TG-SDS buffer for running polyacrylamide gels and then electrotransfer of the proteins on the gel onto membranes. I found out that there had been excessive bubbles formed compared to what I usually experienced. I tried to make my own SDS for the test and the amount of bubbles were much lesser.
Can anyone explain why there the initial set up resulted in excessive bubbles? Thanks in advance!
Excessive bubbles where? When you are running the gel? Look at your current, and try lowering it. If your TG-SDS buffer has a higher content of salt than you usually use, you can expect a larger current at the same voltage. Try turning it down and running it for a bit longer. I used to run some gels for a very long time 12 hours or so at very low voltages of about 5 volts in the cold room to get very good resolution. Also, if you apply less heat, and are running a non dentatured sample, you will get less denaturation by running your gel in the cold room.
